how can i set defense units?

For an offline setup, mainly you will want to pick 1 flank, left or right, to defend, and set it up between 70%  to 100% melee, just depending on where you currently are in the game. 70% will have 2 arrow slits, 2 quicklime/lime powder bombs, 80% will have 1 arrow slit, 3quicklime/lime powder bombs and 100% melee will have 4 quicklime/lime powder bombs. You will also want firemoat.
